dialogue entre préformation et épigenèse est au cœur de la vie. Ce
qui est transmis de façon héréditaire et stable, est une règle de
mise en place. Cette règle a la particularité d'offrir des choix
multiples. Cela se remarque aussi bien dans le système nerveux,
que le système circulatoire ou le système immunitaire.
L'interaction avec l'environnement (qui ne peut être le même
pour deux individus), conduit à la sélection, par stabilisation
(simple pérennité) d'une partie de ce qui a été offert
initialement. On a là la base de l'apprentissage, et de la genèse
de l'identité. |
 The
dialogue between preformation and epigenesis is at the core of
life. A construction rule is what is transmitted through heredity.
This rule offers multiple choices. This can be seen most easily in
the nervous system, the blood system or the immune system. The
interaction with the environment (which cannot be the same for two
individuals), leads to the selection, by a stabilisation process
(ensuring perennity) of a fraction of what has been offered
initially as choices. This is the basis for learning, as well as
for the generation of identity. |

le XVIIIe siècle la philosophie a été envahie par un
"biologisme" mécaniste. La Mettrie en est l'un des meilleurs
exemples. D'où la tentation de parler de l'automate cérébral comme
d'un automate (servo-)mécanique. La science du calcul, qui est en
révolution depuis le début des années 1930 ne s'accomode pas de la
mécanique, même de sa forme élaborée en servo-mécanisme. Il existe
en effet un rôle essentiel du lieu du calcul dans son
développement. Si, donc, on devait encore parler d'automate
cérébral, il serait essentiel de le placer dans le lieu où il
apprend sans cesse, dans son environnement. En quelque sorte les
approches kantienne doivent s'incarner pour prendre en compte la
réalité de l'objet qu'est le cerveau. |
Since
the time of the "Enlightenment", philosophy has been continuously
invaded by a "mechanistic biologism". Julien Offroy de la Mettrie
is one of the best examples of this trend. Hence the images of the
cerebral automaton as a (servo-)mechanism
(controlled mainly by cybernetic feedback processes). On the other
hand, computer science, which has been undergoing a revolution
since the early 1930s, cannot accommodate mechanics, even
considering servo-mechanism. It is indeed immersed in a
development where the situation of the calculation in time and
space plays a crucial role. So if we were still to speak of a
cerebral automaton, it would be essential to put it in the place
where it learns, in its environment. In a certain way, the Kantian
views must be embodied, to take into account the brain object. |

spite of their conceptual difficulty (the human mind tends to look
first for an external driving principle in all things), selective
theories provide remarkably powerful explanations. The principle
of stability in time provides them with an exceptional power of
creation. The work published here combine several approaches
discussed during a weekly seminar held at the Institut de Biologie
Physico-Chimique on wednesday afternoons from 1971 to 1977.
Application of the principle to the immune response, in
collaboration with Gabriel Ruget, led to the design of a first
model for neuronal networks. |

The
conceptual consequences of the principle of selective
stabilization of neuronal networks ask questions about the nature
of language. A deep conjecture about its consequences needs
to be explored, around the concept of homeostasis. It starts from
the very simple idea that strength comes from weakness: it is
because neuronal networks cannot be exactly programmed to make
accurate homeostats that they lead to repeats of approximate
structures that are quasi-homeostatic. Such networks have the
interesting property that they can trigger an automatic scanning
process of the network. Why not propose this at the origin of
consciousness, once the linguistic structures have appeared,
perhaps as the consequence of another approximation, a duplication
of a primitive structure. This was the root of a dialog between
biology and formal properties of syntactic structures proposed by
Noam Chomsky. |

mort, j'ai montré que l'épigraphe du livre célèbre de Jacques
Monod, Le Hasard et la Nécessité, la citation qu'il
attribue à Démocrite ne peut être
correcte, mais que, bien au contraire, la pensée Grecque supposait
le règne de l'ordre, et que les choses évoluent par l'exploration.
Je montrais de plus que le concept d'entropie était associée de
façon erronée au désordre, alors que, là encore, il aurait fallu
l'associer à l'exploration. Quelques conséquences biologiques de
ces observations étaient aussi discutées. |
In
this lecture in memory of the tenth anniversary of his death, I
showed that the epigraph of the famous book of Jacques Monod, Chance
and Necessity, the quotation he attributes to Democritus
cannot be correct, but that, in contrast, there was rule of order
in Greek thought that allowed things to evolve through
exploration. I further argued that the concept of entropy was
wrongly associated to disorder, instead of, once again
exploration. Some biological consequences of these views were also
discussed. |

Scientist, under the revealing label 'French symbolism'! To
check the incredible stupidity of the comment, please read the
book and test your intellectual background; the improbable author
of the book review certainly did not. As an example (but all the
review goes along many similar totally lazy illiterate readings)
the reviewer ascribes to the author of the book the contention
that "The concept of energy, [the author] curiously
ascribes to Mesmer...", which, of course, is a totally fanciful
view of what is in the book... The rest is even more comical.
Reviewers should have some general intellectual education (and be
knowledgeable in domains other than their strict technical
competence) and read first, before reviewing, and should not
invent data as irreal as the "arms of mass destruction" in Iraq!]

Creation postdates the World. Among the most recent discoveries
in the conceptualisation of the World (Reality) the idea of
recursivity, within the simplest objects, those of arithmetics,
suggests a path to follow. Would not it be that path that
matters follows when it becomes Life? This is the core of the
question within our most recent approaches, and the very driving
principle that led us to try and understand the nature of

his inaugural lesson at the Collège de France, François Jacob
reflected about possibly analogies between the structure of the
written language and that of the genetic programme. A few years
later he discussed the topic with the linguist Roman Jakobson.
This theme has initiated my experimental work on the role of
"secundary punctuation" in the process of translation of the
genetic message. This is discussed in this text of the journal Change
(Seghers), after I had uncovered an
unexpected function of secundary punctuation in bacteria. |

M Porcar, A Danchin, V de Lorenzo Confidence, tolerance, and allowance in biological engineering: The nuts and bolts of living things BioEssays (2015) 37: 95-102 Abstract The emphasis of systems and synthetic biology on quantitative understanding of biological objects and their eventual re-design has raised the question of whether description and construction standards that are commonplace in electric and mechanical engineering are applicable to live systems. The tuning of genetic devices to deliver a given activity is generally context-dependent, thereby undermining the re-usability of parts, and predictability of function, necessary for manufacturing new biological objects. Tolerance (acceptable limits within the unavoidable divergence of a nominal value) and allowance (deviation introduced on purpose for the sake of flexibility and hence modularity, i.e. fitting together with a variety of other components) are key aspects of standardization that need to be brought to biological design. These should endow functional building blocks with a pre-specified level of confidence for bespoke biosystems engineering. However, in the absence of more fundamental knowledge, fine-tuning necessarily relies on evolutionary/combinatorial gravitation toward a fixed objective |

Chemical reactivity drives spatiotemporal organisation of bacterial metabolism FEMS Microbiology Reviews (2015) 39: 96-119 Abstract We examine how bacterial metabolism is shaped by chemical constraints acting on the material and dynamic layout of enzymatic networks and beyond. These are moulded not only for optimisation of given metabolic objectives (e.g. synthesis of a particular amino acid or nucleotide) but also for curbing the detrimental reactivity of chemical intermediates. Besides substrate channelling, toxicity is avoided by barriers to free diffusion (i.e. compartments) that separate otherwise incompatible reactions, along with ways for distinguishing damaging vs. harmless molecules. On the other hand, enzymes age and their operating lifetime must be tuned to upstream and downstream reactions. This time dependence of metabolic pathways creates time-linked information, learning and memory. These features suggest that the physical structure of existing biosystems, from operon assemblies to multicellular development may ultimately stem from the need to restrain chemical damage and limit the waste inherent to basic metabolic functions. This provides a new twist of our comprehension of fundamental biological processes in live systems as well as practical take-home lessons for the forward DNA-based engineering of novel biological objects |

| Avant-Propos: Au moment de la création de l'Institut de Biologie Physico-Chimique, la science, à la fois pour des raisons épistémologiques et technologiques, se trouvait séparée en compartiments artificiels selon une classification hiérarchique résumée au milieu du siècle dernier par Auguste Comte: Mathématiques, Physique, Chimie, Biologie. Mais le développement des idées qui conduisait à une conception unitaire de la science atteignait à cette époque un point culminant. Dans son allocution à l'occasion du 25e anniversaire de la fondation de l'Institut de Biologie Physico-Chimique, André Mayer, Professeur au Collège de France et Directeur du Service de Physiologie, distinguait trois cas où la création d'une institution de recherche est nécessaire: celui où cette institution doit donner refuge à la liberté de recherche menacée par un conformisme imposé, ainsi est né le Collège de France; le second cas est celui où l'on veut rassembler des hommes de science avec des praticiens voués à l'application des découvertes, et ce fut la raison de la création de l'Institut Pasteur; le troisième cas est celui où l'on veut faire vivre et travailler côte à côte des chercheurs de différentes disciplines sur des problèmes aux frontières de ces disciplines, et c'est ce qu'a voulu faire le baron Edmond de Rothschild en fondant l'Institut de Biologie Physico-Chimique. this prescient book Schrödinger postulates that heredity behaves
as if borne by an "aperiodic crystal", thus foreseeing the
discovery of the double helix. This text is however marked by the
ideology of the time (1944), obsessed by the idea of degradation.
This made him see life as a perpetual fight against the increase
of entropy, but he notes that this would not fit with what his
colleagues physicists would think. After all the lowest entropy
would be found in a crystal at zero Kelvin, not quite life
conditions.... |

mutations ("adaptive mutations") in bacteria. This lettre
challenges this interpretation by showing that the experimental
setup is heavily biased. A series of experiments, performed with
students of the Microbiology course at the Institut Pasteur de
Paris later on demonstrated
that bacteria could only be Lamarkian if they were very
intelligent organisms! Modulation of mutagenesis under
stressful conditions is enough to give rise to the behaviour
observed. |

psychoanalytic standard model rests on the Œdipus myth. It rests
on the death of the father. We note that it appeared just before
the largest massacre in the history of Mankind (the Great
War) when the fathers sent their sons to death. Was not Œdipus
himself an abandoned child? We propose in this text a new reading
of the myth, where creation, that of the unexpected, is
systematically rejected, and where the death of the progeny plays
a role at least as important as that of its counterpart, long
considered as the only important one. Who remembers Jephte's

Opening: Imagine the situation: “One day, you notice an old country woman crossing a downtown street, just about to put one foot down on the rails of the streetcar line. […] Suppose, now, that at the very moment she puts her foot on the rail a streetcar comes rushing down the tracks toward her. If the old woman does not notice the car and continues across the tracks nothing will happen. But if someone should happen to shout ‘Look out, old woman!’ what would be her natural reaction? […] she would suddenly become flustered and would pause to decide whether to go on or step back […] the mere words ‘Look out, old woman!’ would be as dangerous a weapon as any knife or firearm. […] the man who sounds the warning actually becomes a murderer!" (Ranpo, 1956). In this sense, the Asilomar conference on genetic engineering in 1974 was the perfect crime in using cautionary exclamations asa weapon that had far-reaching consequences for the emerging field of molecular biology: a moratorium stopped research in some places while others proceeded and took the lead. The same fate may lie in store for synthetic biology (SB). The usual crooners have understood the importance of resorting to appeals to ethics and playing on people’s fears to make themselves known and have begun a discussion of the purported dangers of creating organisms a la carte. Such clamouring for attention may yet end up being a shot to our own feet. Moreover, does minimization of a bacterial genome really pose ethical questions? Should we worry, and if so why? After all, what is the purpose of SB? We need to explore the knowledge that we are required to master to (re)construct life in order to gain a better idea of the economic and ethical questions that we have to ask before we start asking them.
