Sperm motility assay
(V. Orgogozo, 2005)
1. collect flies on a CO2 pad. Put a few males in wells + PBS.
2. hold the fly with a forceps and pull the abdomen tip with the other forceps. The reproductive tissues should come out.
3. take both seminal vesicles and both testis with a forceps and put
everything (PBS+tissues) on a slide. A drop of PBS will go on the slide
with the tissues.
4. cover with a cover slip.
5. look for motile sperm within the next two minutes. Use a good dissecting microscope. I use a Nikon SMZ1500.
