Ovariole counts
(V. Orgogozo, 2005)
1. collect flies on the CO2 pad. Put a few females in wells + PBS.
2. hold the fly with a forceps and pull the abdomen tip with the other forceps. The pair of ovaries should come out.
3. put each ovary in a single well + PBT.
4. use two tungstene needles to separate the ovarioles (from posterior end to anterior).
5. count ovarioles once they are all very well separated.
Use a good microscope, with transmitted light if possible. I use a Nikon SMZ1500.
