Octanoic acid resistance test
1. sort flies according to genotype on the CO2 pad. Cut the left or
right wing according to genotype using small dissection scissors.
2. put flies in a Petri dish 35x10. Wait at least 20 min until they all recover.
3. prepare a 1cm x 1cm filter paper with 2uL of ocatonic acid 99% on a
Petri dish cover. Leave it for 5min in the Petri dish before using it.
4. place the cover+ filter paper in place of the other cover.
5. record the number of flies every 20 min.
Corbin Jones’ protocol:
using a modified version of Jones(1998) - i use filter paper to prevent
accidental drowning in the toxin and a greater (10X) dose of
toxin. All expts are conducted at 25 C at 65% relative
humidity. D simulans males survive ~6 minutes; females
survive ~10 minutes. D. sechellia males, in contrast, survive ~45
minutes; D. sechellia female survive >60 minutes on average (and may
survive more than 2 hours).
